Overview

Spexib is an anticancer formulation. It is a prescription medicine used in the treatment of Non-small cell lung cancer (NSCLC). It is available in the form of capsules with a strength of 150mg. This drug belongs to a class of medicines known as protein kinase inhibitors. The Spexib (Ceritinib) capsule works by blocking the activity of abnormal protein that helps the cancer cells to multiply in the body.

Pharmacodynamics

Ceritinib is the active ingredient of Spexib. It works by inhibiting Anaplastic lymphoma kinase (ALK) which is also called ALK tyrosine kinase receptor or simply CD246 (cluster of differentiation 246). Nearly 4-5% of NSCLCs comprise a chromosomal rearrangement that is responsible for the generation of a fusion gene between EML4 (echinoderm microtubule-associated protein-like 4) and ALK (anaplastic lymphoma kinase), leading to constitutive kinase activity that further contributes to carcinogenesis and is thought to drive the malignant phenotype. The compound exerts its therapeutic effect by inhibiting autophosphorylation of ALK, ALK-mediated phosphorylation of the downstream signaling protein STAT3, and proliferation of ALK-dependent cancer cells. Ceritinib inhibited clinical proliferation of cell lines expressing EML4-ALK and NPM-ALK fusion proteins and showed dose-dependent inhibition of EML4-ALK-positive NSCLC xenograft growth in mice and rats.
